It's just a coincidence. The team's been at their Enstone facility since the early 90's and this is the 21st car built at their Enstone facility, hence E21.

This happens from time to time. This year, the Sauber team named their car the C32...Mercedes C32 and in 2011 Ferrari tried to name their car the F150 to celebrate Italy's 150 year anniversary of independance but of course they were threatened with a lawsuit from Ford so they changed the name.
